本文目的是用免疫组织化学方法显示催产素在卵泡壁细胞中和幼稚黄体中的定位.实验用3头性周期正常的母牛.分别于性周期的第6天和7~17天之间以及性周期末摘取卵巢.于显微镜下确定黄体.将组织块分别固定于Bouin氏液及戊二醛苦味酸及冰醋酸混合液(GPA)中6或24小时.固定后常规石蜡包埋,切片厚5μm用于免疫细胞化学染色.相邻切片HE染色.切片按Sternberger(1979)间接过氧化物酶-抗过氧化物酶(PAP)法进行免疫细胞化学染色.第一抗体用兔抗合成的催产素抗血清,稀释度为1:500.第二抗体用驴抗兔抗血清.
